Light intensity significantly influences the novel regulatory function of LiMYB108, a MYB family transcription factor, in floral fragrance. However, the precise way in which light's intensity impacts the release of floral scents is uncertain. Nuclear localization and light-intensity-dependent expression characterize the R2R3-type MYB transcription factor LiMYB108, which was isolated in this study. Light intensities of 200 and 600 mol m⁻¹ s⁻¹ considerably influenced the expression of LiMYB108, consistent with the escalating production of monoterpenes under the same light regime. In Lilium, VIGS-mediated LiMYB108 silencing not only hampered the formation of ocimene and linalool, but also suppressed the expression of LoTPS1; in contrast, transiently increasing LiMYB108 levels produced the opposite consequences. Yeast one-hybrid assays, coupled with dual-luciferase assays and electrophoretic mobility shift assays (EMSA), proved that LiMYB108 directly promoted the expression of LoTPS1 by binding to the MYB binding site (MBS), a sequence of CAGTTG. Light intensity's effect was clearly seen in the high expression levels of LiMYB108, a transcription factor that activated the expression of LoTPS1, and thus facilitated the production of ocimene and linalool, key components in floral fragrance. Within CG (mCG) sequence contexts, DNA methylation, displaying transgenerational consistency and a high epimutation rate, can yield genealogical information at short intervals. Despite the existence of meta-stability and the generation of mCG variants through mechanisms unrelated to epimutation, such as exposure to environmental stresses, the ability of mCG to capture genealogical data at micro-evolutionary scales is unclear. We characterized the DNA methylation variation within accessions of the geographically widespread apomictic dandelion (Taraxacum officinale), contrasting the impact of distinct light conditions applied in a controlled experimental setup. By utilizing a reduced-representation bisulfite sequencing approach, we find that light exposure resulted in the induction of differentially methylated cytosines (DMCs) in all sequence contexts, exhibiting a strong bias toward transposable elements. CG context DMCs were the primary cause of the disparities in accessions. Hierarchical clustering of samples, employing total mCG profiles as the basis, perfectly segregated samples by accession identity, with light conditions playing no role. Based on microsatellite markers as a benchmark of genetic separation within the clonal lineage, we establish a strong relationship between genetic divergence in accessions and overall methylation patterns (mCG). Metabolic syndrome or not, bariatric surgery has consistently proven to be the most effective treatment for obesity. One anastomosis gastric bypass (OAGB), a well-established bariatric procedure, has yielded excellent outcomes over the past two decades of development. The single anastomosis sleeve ileal (SASI) bypass is a newly developed, innovative bariatric and metabolic surgical technique. A parallel can be drawn between the execution of these two tasks. In this study, we present our SASI procedure, building upon the historical experience of the OAGB at our center.
From March 2021 to June 2022, thirty obese patients underwent SASI surgery. We present, step-by-step, our OAGB techniques in this demonstration, and key learnings from our actual experience (as shown in the video), which lead to satisfactory surgical outcomes. Open surgery was not required in any instance. In terms of operative time, blood loss, and hospital stay, the mean values were 1352 minutes, plus or minus 392 minutes; 165 milliliters, plus or minus 62 milliliters; and 36 days, plus or minus 8 days, respectively. The postoperative period was uneventful, with no leakage, bleeding, or mortality observed. At six months, the total weight loss percentage was quantified at 312.65%, while the excess weight loss percentage was 753.149%. Improvements in type 2 diabetes (11/11, 100%), hypertension (14/26, 538%), dyslipidemia (16/21, 762%), and obstructive sleep apnea (9/11, 818%) were evident six months following surgery.
Our practical experience with the SASI technique underscored its viability and potential support for surgeons in performing this promising bariatric procedure with minimal complications.

From the 87 patient-related adverse events, the most frequent was perforation (19, 21.84%), followed by a device becoming embedded in the tissue or plaque (10, 11.49%), and abdominal pain (8, 9.20%). Two of the 19 patients who experienced perforation necessitated open surgical repair, and one required repair through a minimally invasive laparoscopic procedure.
